  Our purpose in this study was to present a method for estimating the specific loss power (SLP) in magnetic hyperthermia in the presence of an external static magnetic field (SMF) and to investigate the SLP values estimated by this method under various diameters (
  <em>D</em>) of magnetic nanoparticles (MNPs) and amplitudes (
  <em>H<sub>0</sub></em>) and frequencies (
  <em>f</em>) of an alternating magnetic field (AMF). In our method, the SLP was calculated by solving the magnetization relaxation equation of Shliomis numerically, in which the magnetic field strength at time 
  <em>t </em>(
  <em>H</em>(
  <em>t</em>)) was assumed to be given by 
  <img src="Edit_6fb383d3-55ee-415f-88de-45435ab921f0.bmp" alt="" />, with 
  <em>H<sub>s</sub></em> being the strength of the SMF. We also investigated the SLP values in the case when the SMF with a field-free point (FFP) generated by two solenoid coils was used. The SLP value in the quasi steady state (
  <em>SLP<sub>qss</sub></em>) decreased with increasing 
  <em>H<sub>s</sub></em>. The plot of the 
  <em>SLP<sub>qss</sub></em> values against the position from the FFP became narrow as the gradient strength of the SMF (
  <em>G<sub>s</sub></em>) increased. Conversely, it became broad as 
  <em>G</em>
  <sub><em>s</em></sub> decreased. These results suggest that the temperature rise and the area of local heating in magnetic hyperthermia can be controlled by varying the 
  <em>H<sub>s</sub></em> and 
  <em>G<sub>s</sub></em> values, respectively. In conclusion, our method will be useful for estimating the SLP in the presence of both the AMF and SMF and for designing an effective local heating system for magnetic hyperthermia in order to reduce the risk of overheating surrounding healthy tissues.

Introduction</title><p>Hyperthermia is one of the promising approaches to cancer therapy [<xref ref-type="bibr" rid="scirp.72330-ref1">1</xref>] [<xref ref-type="bibr" rid="scirp.72330-ref2">2</xref>] . Hyperthermia with use of magnetic nanoparticles (MNPs) (magnetic hyperthermia) was developed in the 1950s [<xref ref-type="bibr" rid="scirp.72330-ref3">3</xref>] and is still under development in the effort to overcome the drawbacks in other hyperthermia modalities such as radiofrequency (RF)-capacitive hyperthermia and ultrasound hyperthermia [<xref ref-type="bibr" rid="scirp.72330-ref4">4</xref>] [<xref ref-type="bibr" rid="scirp.72330-ref5">5</xref>] .</p><p>MNPs generate heat in an alternating magnetic field (AMF) as a result of hysteresis and relaxational losses, which results in heating of the tissue in which MNPs accumulate [<xref ref-type="bibr" rid="scirp.72330-ref6">6</xref>] . For small MNPs, the relaxational losses caused by a delay in magnetization relaxation are dominant for heat dissipation [<xref ref-type="bibr" rid="scirp.72330-ref6">6</xref>] . With the development of precise methods for synthesizing functionalized MNPs [<xref ref-type="bibr" rid="scirp.72330-ref7">7</xref>] , MNPs with functionalized surfaces, which have high specificity for tumor tissue, have been developed as heating elements for magnetic hyperthermia [<xref ref-type="bibr" rid="scirp.72330-ref8">8</xref>] . Recently, MNPs with a higher heating efficiency, i.e., specific loss power (SLP) have also been actively developed [<xref ref-type="bibr" rid="scirp.72330-ref9">9</xref>] . Furthermore, there is renewed interest in magnetic hyperthermia as a treatment modality for cancer, especially when it is combined with other, more traditional therapeutic approaches such as the co-delivery of anticancer drugs [<xref ref-type="bibr" rid="scirp.72330-ref10">10</xref>] or radiation therapy [<xref ref-type="bibr" rid="scirp.72330-ref11">11</xref>] . From these aspects, magnetic hyperthermia has received much recent attention.</p><p>The estimation of SLP is important for evaluating the heating efficiency of MNPs, for optimizing the parameters of AMF, and for the optimal design of MNPs in an attempt to establish the effectiveness of magnetic hyperthermia. Rosensweig’s model [<xref ref-type="bibr" rid="scirp.72330-ref6">6</xref>] has often been used for the estimation of SLP. His model, however, is based on the so-called linear magnetization assumption [<xref ref-type="bibr" rid="scirp.72330-ref12">12</xref>] , and thus it is said that his model is strictly valid only in the limit of small amplitude and frequency of AMF. Recently, we have presented three methods for estimating SLP and compared the SLP values estimated by the three methods under various conditions of MNPs and AMF [<xref ref-type="bibr" rid="scirp.72330-ref13">13</xref>] . Furthermore, we investigated the validity of Rosensweig’s model in comparison with the numerical solution of the magnetization relaxation equation of Shliomis [<xref ref-type="bibr" rid="scirp.72330-ref13">13</xref>] [<xref ref-type="bibr" rid="scirp.72330-ref14">14</xref>] .</p><p>It is also important to heat the targeted tumor to the desired temperature without damaging the surrounding healthy tissues in order to enhance the effectiveness of magnetic hyperthermia. In 2009, Tasci et al. [<xref ref-type="bibr" rid="scirp.72330-ref15">15</xref>] proposed and designed a system that focuses the heat into very small regions using a static magnetic field (SMF) with a field- free point (FFP) generated by two solenoid coils, and reported that this method will be useful for making magnetic hyperthermia a more effective approach to cancer therapy with a decreased risk of heating surrounding healthy tissues. We have also investigated the usefulness of this method using phantom experiments and reported that it is useful for controlling the temperature rise in magnetic hyperthermia [<xref ref-type="bibr" rid="scirp.72330-ref16">16</xref>] . Although this approach appears to be useful for focusing the heat deposition into the targeted tissue or organ, it would be necessary to investigate the effectiveness of this approach under various conditions of MNPs, AMF, and SMF. Furthermore, it would be desired to derive an equation describing the energy dissipation of MNPs in the presence of both the AMF and SMF for estimating and controlling the heat deposition into the targeted tissue or organ and for designing an effective local heating system.</p><p>Our purpose in this study was to present a method for estimating the SLP in magnetic hyperthermia in the presence of an external SMF and to investigate the SLP values estimated by this method under various conditions of MNPs, AMF, and SMF.</p></sec><sec id="s2"><title>2. Materials and Methods</title><sec id="s2_1"><title>2.1. Theory</title><p>The magnetization relaxation equation of Shliomis [<xref ref-type="bibr" rid="scirp.72330-ref12">12</xref>] [<xref ref-type="bibr" rid="scirp.72330-ref14">14</xref>] is given by</p><disp-formula id="scirp.72330-formula209"><label>, (1)</label><graphic position="anchor" xlink:href="http://html.scirp.org/file/3-2310677x3.png"  xlink:type="simple"/></disp-formula><p>where M is the magnetization of MNPs under the magnetic field H, Ω is the flow velocity, f is the volume fraction, and η is the viscosity of the suspending fluid. When there is no bulk flow and M and H are collinear, Equation (1) is reduced to the following equation [<xref ref-type="bibr" rid="scirp.72330-ref6">6</xref>] :</p><disp-formula id="scirp.72330-formula210"><label>. V<sub>H</sub> is taken as the hydrodynamic volume of MNP that is larger than the magnetic volume <inline-formula><inline-graphic xlink:href="http://html.scirp.org/file/3-2310677x8.png" xlink:type="simple"/></inline-formula> for MNP of diameter D. Because <inline-formula><inline-graphic xlink:href="http://html.scirp.org/file/3-2310677x32.png" xlink:type="simple"/></inline-formula><inline-formula><inline-graphic xlink:href="http://html.scirp.org/file/3-2310677x33.png" xlink:type="simple"/></inline-formula> must be time-periodic in the steady state, the SLP value for the i-th cycle of the M-H curve (denoted by SLP<sub>i</sub>) can be given by</p><p><inline-formula><inline-graphic xlink:href="http://html.scirp.org/file/3-2310677x34.png" xlink:type="simple"/></inline-formula>.(12)</p><p>It should be noted that when i is sufficiently large, the second term of the right-hand side of Equation (12) can be neglected and SLP<sub>i</sub> approaches the steady state. We denote the SLP<sub>i</sub> value in the quasi steady state by SLP<sub>qss</sub>. Actually, SLP<sub>qss</sub> was taken as the SLP<sub>i</sub> value in the case when the relative error (RE) was less than 10<sup>−10</sup>. The RE was defined by</p><disp-formula id="scirp.72330-formula220"><label>, (13)</label><graphic position="anchor" xlink:href="http://html.scirp.org/file/3-2310677x35.png"  xlink:type="simple"/></disp-formula><p>where <inline-formula><inline-graphic xlink:href="http://html.scirp.org/file/3-2310677x36.png" xlink:type="simple"/></inline-formula> denotes the absolute value. The integration in Equation (12) was performed by use of the trapezoidal rule [<xref ref-type="bibr" rid="scirp.72330-ref17">17</xref>] (“trapz” in MATLAB&#174;; The MathWorks, Inc., Natick, MA, USA) and the convolution integral was calculated using the MATLAB function (“conv”).</p></sec><sec id="s2_2"><title>2.2. Simulation Studies</title><p>In this study, we assumed that MNPs consisted of two kinds of iron oxide nanoparticles, i.e., maghemite (γ-Fe<sub>2</sub>O<sub>3</sub>) and magnetite (Fe<sub>3</sub>O<sub>4</sub>). We fixed τ<sub>0</sub>, δ, M<sub>d</sub>, K, η, ρ, f, and T to be 10<sup>−</sup><sup>9</sup> s, 2 nm, 414 kA/m, 4.7 kJ/m<sup>3</sup>, 0.00235 kg/m/s, 4600 kg/m<sup>3</sup>, 0.003, and 37˚C, respectively, for maghemite [<xref ref-type="bibr" rid="scirp.72330-ref18">18</xref>] [<xref ref-type="bibr" rid="scirp.72330-ref19">19</xref>] . For magnetite, we fixed τ<sub>0</sub>, δ, M<sub>d</sub>, K, η, ρ, f, and T to be 10<sup>−</sup><sup>9</sup> s, 2 nm, 446 kA/m, 9.0 kJ/m<sup>3</sup>, 0.00235 kg/m/s, 5180 kg/m<sup>3</sup>, 0.003, and 37˚C, respectively [<xref ref-type="bibr" rid="scirp.72330-ref19">19</xref>] . When H<sub>0</sub>, f, and D were fixed, they were taken as 20 mT, 300 kHz, and 20 nm, respectively. It should be noted that the unit of mT can be converted to kA/m by use of the relationship 1 mT = 0.796 kA/m.</p><p>When considering the control of the temperature rise using the SMF with a gradient strength of G<sub>s</sub>, the strength of the SMF at a distance of x from the FFP (H<sub>s</sub>(x)) was given by</p><disp-formula id="scirp.72330-formula221"><label>. (14)</label><graphic position="anchor" xlink:href="http://html.scirp.org/file/3-2310677x37.png"  xlink:type="simple"/></disp-formula></sec></sec><sec id="s3"><title>3. Results</title><p>As shown in Equation (12), the SLP<sub>i</sub> value depends on the cycle number of the M-H curve. Thus, we calculated the SLP<sub>i</sub> value in the quasi steady state, i.e., the SLP<sub>qss</sub> value under the condition in which the RE given by Equation (13) was less than 10<sup>−10</sup>, as previously described. <xref ref-type="fig" rid="fig1">Figure 1</xref>(a) shows the common logarithm of the RE given by Equation (13) as a function of the number of cycles for maghemite, whereas <xref ref-type="fig" rid="fig1">Figure 1</xref>(b) shows the case for magnetite. In these simulations, we neglected the second term in the right-hand side of Equation (12), because it approaches zero in the steady state. The τ value given by Equation (3) was 4.10 &#215; 10<sup>−8</sup> s and 1.71 &#215; 10<sup>−6</sup> s for maghemite and magnetite, respectively. As shown in <xref ref-type="fig" rid="fig1">Figure 1</xref>, the speed with which the SLP<sub>i</sub> value reached the quasi steady state depended on the τ value, i.e., the larger the τ value, the more slowly the SLP<sub>i</sub> value reached the quasi steady state. As shown in <xref ref-type="fig" rid="fig1">Figure 1</xref>(b), there was a tendency for the RE to increase with increasing H<sub>s</sub>. Note that the plateau regions in <xref ref-type="fig" rid="fig1">Figure 1</xref> correspond to the so-called “machine epsilon”.</p><p><xref ref-type="fig" rid="fig2">Figure 2</xref>(a) shows the M-H curves in the quasi steady state calculated from Equation (8) at an f of 300 kHz for maghemite. In this case, H<sub>s</sub> was varied from 0 to 30 mT with steps of 10 mT. For comparison, <xref ref-type="fig" rid="fig2">Figure 2</xref>(b) shows those for magnetite. It should be noted that <inline-formula><inline-graphic xlink:href="http://html.scirp.org/file/3-2310677x38.png" xlink:type="simple"/></inline-formula> was normalized by the saturation magnetization (M<sub>s</sub>) given by<inline-formula><inline-graphic xlink:href="http://html.scirp.org/file/3-2310677x38.png" xlink:type="simple"/></inline-formula><inline-formula><inline-graphic xlink:href="http://html.scirp.org/file/3-2310677x39.png" xlink:type="simple"/></inline-formula>. In these simulations, H<sub>0</sub> was fixed at 20 mT and D was assumed to be 20 nm. <xref ref-type="fig" rid="fig3">Figure 3</xref> shows the case when f was taken as 600 kHz. As shown in <xref ref-type="fig" rid="fig2">Figure 2</xref> and <xref ref-type="fig" rid="fig3">Figure 3</xref>, the area of the M-H curve decreased with increasing H<sub>s</sub> and a large difference in the M-H curve was observed between maghemite and magnetite.</p><p><xref ref-type="fig" rid="fig4">Figure 4</xref>(a) shows the SLP<sub>qss</sub> values calculated from Equation (12) as a function of H<sub>s</sub> with D being varied from 10 nm to 30 nm with steps of 5 nm for maghemite, whereas <xref ref-type="fig" rid="fig4">Figure 4</xref>(b) shows the case for magnetite. In these simulations, H<sub>0</sub> and f were fixed at 20 mT and 300 kHz, respectively. As in <xref ref-type="fig" rid="fig4">Figure 4</xref> and <xref ref-type="fig" rid="fig5">Figure 5</xref>, the SLP<sub>qss</sub> value decreased with increasing H<sub>s</sub> in all cases.</p><p><xref ref-type="fig" rid="fig7">Figure 7</xref>(a) shows the SLP<sub>qss</sub> values calculated from Equation (12) as a function of the distance from the FFP (x) at various values of G<sub>s</sub> (1, 2, 5, and 10 T/m) for maghemite, whereas <xref ref-type="fig" rid="fig7">Figure 7</xref>(b) shows the case for magnetite. In these simulations, H<sub>s</sub> at x, i.e., H<sub>s</sub>(x) was calculated from G<sub>s</sub> and x using Equation (14), and D, H<sub>0</sub>, and f were fixed at 20 nm, 20 mT, and 300 kHz, respectively. Discussion</title><p>In this study, we presented a method for estimating the SLP in magnetic hyperthermia in the presence of both the AMF and SMF, which was derived by solving the magnetization relaxation equation of Shliomis [<xref ref-type="bibr" rid="scirp.72330-ref14">14</xref>] numerically. We also presented the results obtained by simulation studies under various conditions of MNPs, AMF, and SMF. Our results shown in <xref ref-type="fig" rid="fig4">Figure 4</xref> to <xref ref-type="fig" rid="fig7">Figure 7</xref> suggest that the SLP in magnetic hyperthermia can be controlled using the SMF and that our method will be useful for selecting the optimal parameters for controlling the temperature rise in magnetic hyperthermia in order to reduce the risk of overheating surrounding healthy tissues.</p><p>As shown in <xref ref-type="fig" rid="fig1">Figure 1</xref>, the RE given by Equation (13) for maghemite (<xref ref-type="fig" rid="fig1">Figure 1</xref>(a)) became less than the threshold (10<sup>−</sup><sup>10</sup> in this study) faster than that for magnetite (<xref ref-type="fig" rid="fig1">Figure 1</xref>(b)). As previously described, there was a large difference in the τ value given by Equation (3) between maghemite and magnetite (4.10 &#215; 10<sup>−</sup><sup>8</sup> s and 1.71 &#215; 10<sup>−</sup><sup>6</sup> s for maghemite and magnetite, respectively, under the condition described in the “Simulation Studies” section). Thus, the above difference appears to be due to a difference in the τ value. Similarly, a large difference in the M-H curve was observed between maghemite (<xref ref-type="fig" rid="fig2">Figure 2</xref>(a)) and magnetite (<xref ref-type="fig" rid="fig2">Figure 2</xref>(b)). This difference also appears to be due to a difference in the τ value.</p><p>As shown in <xref ref-type="fig" rid="fig2">Figure 2</xref> and <xref ref-type="fig" rid="fig3">Figure 3</xref>, the area of the M-H curve calculated from Equation (8) decreased with increasing H<sub>s</sub>. The area of the M-H curve directly represents the power loss during one cycle of the hysteresis loop. Thus, the above finding corresponds to the fact that SLP<sub>qss</sub> decreases with increasing H<sub>s</sub> (<xref ref-type="fig" rid="fig4">Figure 4</xref> to <xref ref-type="fig" rid="fig6">Figure 6</xref>).</p><p>We previously made an experimental device that allows for magnetic hyperthermia in the presence of the SMF that was generated using a Maxwell coil pair [<xref ref-type="bibr" rid="scirp.72330-ref16">16</xref>] . This device can also generate an FFP and move it by varying the DC electric currents applied to the individual Maxwell coils [<xref ref-type="bibr" rid="scirp.72330-ref16">16</xref>] . We investigated the effect of SMF on the temperature rise in magnetic hyperthermia using this device and then evaluated the feasibility of controlling the temperature rise using an SMF with an FFP under various conditions of the AMF and SMF. Our experimental results demonstrated that the SLP value (referred to as “SAR” in our previous paper [<xref ref-type="bibr" rid="scirp.72330-ref16">16</xref>] ) decreased with increasing H<sub>s</sub> [<xref ref-type="bibr" rid="scirp.72330-ref16">16</xref>] . Thus, the present results shown in <xref ref-type="fig" rid="fig4">Figure 4</xref> to <xref ref-type="fig" rid="fig6">Figure 6</xref> appear to support our experimental results [<xref ref-type="bibr" rid="scirp.72330-ref16">16</xref>] . Furthermore, our experimental results showed that the area of local heating became narrow with increasing G<sub>s</sub>. Conversely, it became broad with decreasing G<sub>s</sub> [<xref ref-type="bibr" rid="scirp.72330-ref16">16</xref>] . As shown in <xref ref-type="fig" rid="fig7">Figure 7</xref>, the plot of the SLP values against the position from the FFP became narrow as G<sub>s</sub> increased, whereas it became broad as G<sub>s</sub> decreased. These results also appear to support our experimental results described above [<xref ref-type="bibr" rid="scirp.72330-ref16">16</xref>] .</p><p>In our previous study [<xref ref-type="bibr" rid="scirp.72330-ref16">16</xref>] , we treated the case in which the FFP is moved in one dimension by use of a single Maxwell coil pair. However, it would be possible to extend it to two or three dimensions by use of multiple Maxwell coil pairs. Theoretically, the magnetic field strength generated by a Maxwell coil pair can be calculated from the Biot-Savart law [<xref ref-type="bibr" rid="scirp.72330-ref20">20</xref>] . Thus, when the electric currents applied to the coils are known, the position of the FFP can be determined by solving the equations describing the magnetic field strength based on the Biot-Savart law [<xref ref-type="bibr" rid="scirp.72330-ref20">20</xref>] . Conversely, when the position of the FFP is given, the electric currents applied to the coils can be determined by the similar procedure. If this could be realized, it would become easy to control the position of the FFP, i.e., the position of local heating. As previously described, the plot of the SLP values against the position from the FFP became broad with decreasing G<sub>s</sub>, whereas it became narrow with increasing G<sub>s</sub> (<xref ref-type="fig" rid="fig7">Figure 7</xref>). These results suggest that the area of local heating can be controlled by varying the value of G<sub>s</sub>. The G<sub>s</sub> value can also be calculated from the Biot-Savart law [<xref ref-type="bibr" rid="scirp.72330-ref20">20</xref>] . Thus, it would become possible to control not only the position but also the area of local heating by controlling the electric currents applied to the coils based on the Biot-Savart law [<xref ref-type="bibr" rid="scirp.72330-ref20">20</xref>] . This will further enhance the feasibility and effectiveness of the magnetic hyperthermia with the temperature rise being controlled for reducing the risk of overheating surrounding healthy tissues.</p><p>In this study, we derived Equation (12) by solving the magnetization relaxation equation of Shliomis [<xref ref-type="bibr" rid="scirp.72330-ref14">14</xref>] (Equation (1)) with an assumption that there is no bulk flow and the magnetization of MNPs and magnetic field are collinear. In this case, Equation (1) is reduced to Equation (2), which can be easily solved using convolution integral as shown in Equation (8). Although Equation (2) appears to be valid in considering the magnetic hyperthermia with small MNPs in the superparamagnetic state and we believe that this study will provide the basis for establishing the effectiveness of such magnetic hyperthermia, it will be necessary to solve Equation (1) without any assumptions or another magnetization equation derived microscopically from the Fokker-Planck equation [<xref ref-type="bibr" rid="scirp.72330-ref12">12</xref>] for more detailed analysis. These studies are currently in progress. As previously described, we targeted the MNPs consisting of maghemite and magnetite with the magnetic and physical properties described in the “Simulation Studies” section. We will also perform further studies for the MNPs with other magnetic and physical properties and/or other MNPs.</p><p>Recently, Dhavalikar et al. [<xref ref-type="bibr" rid="scirp.72330-ref21">21</xref>] have theoretically investigated the feasibility of spatially-focused heating of MNPs guided by the SMF with a field-free region such as the FFP. In their study, the phenomenological magnetization equation derived by Martsenyuk et al. [<xref ref-type="bibr" rid="scirp.72330-ref22">22</xref>] was used instead of the Shliomis’ equation [<xref ref-type="bibr" rid="scirp.72330-ref14">14</xref>] . A comparative study of the present results and those based on the equation of Martsenyuk et al. [<xref ref-type="bibr" rid="scirp.72330-ref22">22</xref>] is also currently in progress.</p></sec><sec id="s5"><title>5. Conclusion</title><p>We presented a method for estimating the SLP in magnetic hyperthermia in the presence of an external SMF, which is based on the numerical solution of the magnetization relaxation equation of Shliomis. We also presented the SLP values estimated under various conditions of MNPs, AMF, and SMF.
