ABSTRACT: This study aimed to optimize serum concentration, synchronization duration, and cell confluence to improve cell-cycle synchronization efficiency in Thanh Chương buffalo fibroblasts. The proportion of nucleated fibroblasts in the G0/G1 phase was the highest at a serum concentration of 0.3% (81.26%) and the lowest in the control group (61.44%), with the difference being statistically significant (P 0.05). The proportion of fibroblasts in the G0/G1 phase was significantly higher in the 96-hour and 120-hour synchronization groups compared with the 24-hour, 48-hour, and 72-hour groups (P 0.05). Similarly, fibroblasts at 80% and 100% confluence exhibited a significantly higher proportion in the G0/G1 phase compared with those at 60% confluence (86.32% and 85.91% vs 70.56%, respectively; P
