  ABSTRACT: As a key process in maintaining cellular metabolic homeostasis, the targeted regulation of autophagy offers new insights into the treatment of metabolic disorders such as diabetes, obesity, and non-alcoholic fatty liver disease (NAFLD). This systematic review examines core strategies for regulating autophagy to improve metabolic disorders, including lifestyle interventions, pharmacological modulation and the use of natural compounds. It also analyses the scientific basis and clinical limitations of existing therapies and explores future directions in tissue-specific drug development, precision medicine applications and interdisciplinary technological integration. The aim is to provide a theoretical foundation and practical reference for the development of safer and more effective metabolic disease treatments, facilitating a shift from broad-spectrum intervention to precise modulation.
