Neural Stem Cells Transplantation Promotes Brain Repair after Chronic Experimental Allergic Encephalomyelitis in Rat

ABSTRACT: The recent studies have showed neural stem cells
(NSCs) transplantation promoted repair and functional recovery of chronic
experimental allergic encephalomyelitis (EAE) animal model for MS based on the
concept of cell replacement. In the present study, NSCs were transplantated
into each lateral ventricle of rats at 10 days and sacrificed at 60 days after
EAE immunization. We found that NSCs transplantation significantly reduced the
clinical signs, suppressed brain inflammation and improved density of myelin. Transplanted
NSCs differentiated into neurons and oligodendrocytes. Moreover, the number of
endogenous neurons and oligodendrocytes in the rats treated with NSCs was
significantly higher compared to the rats with saline. These results suggest an
important neuroprotective role for NSCs that can potentially be exploited in
cell-based therapeutic approaches for EAE.
